The HUMIMIC Chip4 is a cutting-edge multi-organ-on-a-chip platform allowing the simultaneous culture of up to five tissue types. This chip is composed two microfluidic circuits separated by a porous membrane. It provides a more realistic and dynamic model of human physiology. This multi-organ platform enables the modeling of a "body-on-chip" to investigate drug metabolism and diseases in a controlled environment.
General
The HUMIMIC Chip4 allows the co-culture of up to 5 different tissues to model complex organ interactions. The Chip4 is composed of two microfluidic circuits, which are separated by a polycarbonate porouse membrane (1µm pore size). This can be useful to model some biological processes occuring in the organism like the excretion and reabsorption of substances in a kidney model for example.
The HUMIMIC Chips are operated via the HUMIMIC Starter, which can control several chips and chip types at the same time. Pressure and vacuum can be applied and allow the modeling of a lifelike supply of the tissues and also the modulation of shear stress according to the experimental needs of the user. The HUMIMIC Chip2 contains 3 pump membranes (500 µm thick) to control the liquid displacement in the microfluidic circuit, through valves activated by the pressure applied by the HUMIMIC Starter.
The flexibility of the HUMIMIC device allows to study any organ and also biological barrier models like the intestine, the lung or the skin. It is compatible with cell line, primary cells but also biopsies, tissue sections or spheroids. It is also possible to use commercially available culture inserts or even matrix-suported cultures to mimick the 3D environment of the organs. These chips, made of polycarbonate, PDMS and glass, have been designed for long-term experiments allowing the study of disease models and drug's efficacy and toxicity.
The Chip4 device presented here displays two types of cell culture compartments:
- 4x Threadfit 01 (96-well type) - compatible with Transwell inserts or serving as medium reservoir of up to 350 µL.
- 1x Threadfit 05 (24-well type) - compatible with Millicell inserts
This multi-organ-chip is particularly interesting for the prediction of ADMET profiles (adsorption, distribution, metabolisme and excretion) for drug development.
Each chip is provided with the relevant culture compartments which are screwed into the device thanks to a hexagonal socket wrench. For media exchange, the compartments of one circuit need to be opened separately.
The types of culture compartment proposed for each chip are described in the specification section. Note that the nature of the culture compartment can be changed according to what is described in the product catalog of the Documentation section.
All chips are provided prefilled with DPBS supplemented with 1% Penicillin/Streptomycin (PS) but can alos be provided with a mix of 0.01% Gentamicin / 0.1% Amphotericin B (GA) or without any antibiotics (WO) added in the DPBS. For any requests regarding customization, please contact us.
