General
The Akura™ PLUS Hanging Drop System is a 2-plate system: one Akura™ PLUS Hanging Drop plate and one Akura™ 96 Spheroid Microplate. It allows the robust formation of spheroids, and their long-term growth, culturing, and analysis thanks to available protocols.
The formation of spheroids starts with the delivery of a cell suspension (40 µL) in the SureDrop™ inlet funnels of the PLUS plate with a pipette or automated liquid handler. Hanging drops will then form at the bottom and spheroids will be formed by gravity-assisted assembly within 2-4 days.
During spheroid formation, the initial cell suspension should be homogenous for getting reproducible results. To prevent the evaporation of hanging drops, a humidifier pad is included in the system and should be put in the bottom tray of the PLUS plate to ensure optimal humidity.
Once spheroids are constituted, they can be transferred to the Akura™ 96 plate for culturing or downstream analysis. This 96-well plate presents a unique SureXchange well design which allows near complete medium exchange without spheroid loss. These plates are compatible with most laboratory equipment thanks to their ANSI/SLAS format, a 1 mm-observation window for easy imaging, and simple medium collection for biochemical assays.
