The Akura™ 384-well plate facilitates the generation, long-term cultivation, and analysis of 3D cell models. This simple and flexible platform is also compatible with automation systems. The unique well design ensures high reproducibility, scalability, and near-complete medium exchange without spheroid loss. The ImagePro plate is recommended for high-content imaging applications.
General
This 384-well plate enables scaffold-free aggregation and long-term culturing of 3D cell models while being compatible with automation systems for high-throughput experiments. The plate supports several 3D cell models such as spheroids, organoids, and microtissues of up to 750 µm. Cell attachment to the plate's surface is prevented thanks to an ultra-low attachment (ULA) coating. Use of this plate is recommended with cell types known to readily form spheroids in ULA conditions.
Spheroid formation and maturation appear within 2-5 days of incubation at 37°C following cell seeding and brief centrifugation (recommended to remove air bubbles). The maturation process can be improved by tilting the plate to 30° or placing it on the Akura™ Tilting Stand.
The SureXchange unique well design has been developed to ensure near-complete (>90%) medium exchange without spheroid loss due to unintended aspiration. In the 384 plates, the design of the well is acentric. It is thus important to identify where are located the spheroid chamber and the ledge to orient the plate accordingly.
This technology shows great reproducibility and uniformity in spheroid formation and allows for long-term culturing. It also helps improve washing steps to remove unwanted components (medium components, hormones) which could interfere with endpoint assays or spheroid behavior.
With an ANSI/SLAS standard format, this plate is compatible with all existing laboratory equipment, including multi-channel pipettes and automated liquid handling systems for higher throughput. A wide range of readouts can be achieved such as biochemical assays, inverse bright field or fluorescence imaging.
The plate is composed of a black-walled Polystyrene body bonded to a transparent continuous membrane. The black-walled wells help to eliminate fluorescent crosstalk between wells. In the Akura™ 384 Spheroid Microplate, the membrane is made of polystyrene and is 0.125 mm thick. The Akura™ 384 Image Pro results in enhanced imaging quality and depth thanks to its highly transparent 0.025 mm-thick FEP (fluorinated ethylene propylene) membrane.
